For example, if I buy something that's worth $5 and I pay $20, do I ask do you have change for $20? Or do you have change for $15? I heard both but which one is right?

You should say "do you have change for twenty?"

the seller not necessary has change.  there is possibility that on that particular day everyone pays with 20 and the seller uses up all the changes.
